Basic Guidelines for Plant Tissue Culture Lab

If you are novice in the field of Plant Tissue Culture (PTC), then here you can learn the basic guidelines that are necessary for you when you enter a lab.

Plant Tissue Culture lab is used for culturing or growing plants in the labs. Plants can be grown in various ways. However, if you want to go to any such lab or are working in such labs, then you must know few basic things.

I tried hard but could not come across any such particular guidelines on net. So, I try to give you some of the basic guidelines which I have learned from my experience. Since, without them you would be a total illiterate in the plant tissue culturing field.

The most important things you should know are:

  1. You must know botanical names of plants. Try to remember as many plants as much you can.
  2. You must know in brief the anatomy of plants. For example: leaf, stem, etc.
  3. You must know the basic requirements of a plant for its survival
  4. The basic concept of sterilization and various sterilization methods used in lab must be known to you
  5. The common techniques used in plant tissue culturing must be known to you. They are as follows:
    • Micro propagation
    • Haploid culture
    • Embryo culture
    • Somatic embryogenesis
    • Cell suspension culture
    • Protoplast fusion
  6. Try to learn common instruments and chemicals found in a plant tissue culture lab. For example: centrifuge, pH meter, autoclave, salts, etc.
  7. Some common terms like totipotency, embryo rescue, protocorm, etc. must be known to you.
  8. If possible, always know few practical examples where plant tissue culturing techniques are used for plant cultivation. At least, one example for each technique you must remember.
  9. Also try to learn, which methods are employed to modify the characteristics of the plants. For example: Insertion of DNA through a vector, exposure to mutagen, etc.
  10. Last of all, it would be an added advantage if you know some recent developments in this field.

This will help you to handle any plant tissue culture lab.

    I am sorry for late reply.

    Your answer is bit vague for me and would prefer a more nuanced question.

    Howeve, if you are interested in setting up and plant tissue culture lab then first determine what kind of work you plan to do?

    Because if it is simple culturing some basic instruments like LAF, AC, Light source, etc would do.
    But If looking for more extensive work then requirements can vary according to your requirement.

    For example: for analysis of certain protein in your culture you might need electrophoretic unit, detector etc.

    It all depends what you wanna do.

    In general if you talk about rooms, it consists of:

    1) Incubation room
    2) culture room
    3) media preparation room
    4) cleaning room
    in some cases,
    5) analysis room
